We would like imaging collagen (not elastin) within the rat heart (cryocuts) using a 2-photon microscope ( Zeiss LSM 510 equipped with a LASER Coherent Verdi). Is there a good reference/tutorials that can help me going trough this? Suitable excitation wavelenght, SHG, thickness of the cuts, known excitation/emission spectra etc.
